Abstract:
Obesity and its adverse metabolic consequences are major and growing public health challenges. Through the study of individuals with extreme early onset phenotypes such as obesity or insulin resistance we have discovered many previously unrecognised genetic defects. By linking gene discovery with biochemical and in vivo phenotyping our work has provided new information about the normal control of body weight and insulin sensitivity, brought an end to a diagnostic odyssey for many patients and in some instances led to targeted therapies.
